The scientists at Intact Genomics are the premier experts in competent cell production, with unmatched quality, service and the greatest value in the industry. IG custom competent cells are available in chemically competent or electrocompetent format.  You may choose from a variety of common E. coli strains (5-alpha, DH10B, BL21(DE3), etc., or you may simply send us your own strain. Upon request, we are also able to safely store your strain to make production of additional competent cells at a later date convenient and fast.

Intact Genomics uses proprietary methods and technologies to produce the highest transformation efficiencies possible (> 5 x 10^10 cfu/µg DNA).  Using our unique production methods, we have significant experience providing custom competent cells for multiple customers in the pharmaceutical, agricultural and biotech industries allowing significant progress with their research.

    Endonuclease IV (Nfo)
    Endonuclease IV (Nfo) from Escherichia coli is a 32-kD metalloprotein that aids in the repair of damaged DNA. The enzyme functions both as an apurinic/apyrimidinic nuclease (1) and as a 3′-terminal di-esterase (1-4). Its 3′-terminal di-esterase activity is important in the repair of DNA strand breaks generated by oxidation and ionic radiation (2, 3). In such events, the strand breaks terminate with either a 3′ phosphate or a deoxyribose fragment, preventing repair by DNA polymerase I or DNA ligase. Endonuclease IV removes the blocking groups, leaving a free 3′-hydroxyl terminus. This enzyme does not have detectable associated exonuclease or DNA N-glycosylase activity (1).

    T7 Endonuclease I
    T7 Endonuclease I, a stable homodimer of identical 149 amino acid subunits (1) is the product of a recombinant gene in E. coli . Mutations generated by CRISPR/ZFN/TALEN at desired target sites can be PCR-amplified, and the PCR products can be denatured and re-annealed to form mismatched DNA. If the mismatched DNA length position is more than 1 bp, T7 Endonuclease I can recognize and cleave it. It is useful for quantitatively estimating the nuclease-induced mutation frequency of gene edited cells.

    HB101 Chemically Competent Cells
    Intact Genomics (ig®) HB101 chemically competent E. coli cells are suitable for high-efficiency transformation in a wide variety of applications such as cloning and sub-cloning. E. coli HB101 is a K12 x B hybrid strain, containing the recA13 mutation that minimizes recombination and helps insert stability. In addition, it carries the hsdS20(rB-mB-) restriction minus genotype which prevents cleavage of cloned DNA by endogenous restriction enzymes.     Ar.A4 Agrobacterium Chemically Competent Cells
    Transformation Protocol Use this procedure to transform Ar.A4 Chemically Competent Agrobacterium cells. Do not use these cells for electrocompetent transformation.
    1. Place microcentrifuge tubes on ice.
    2. Remove competent cells from the -80 °C freezer and thaw completely on wet ice (10-15 minutes).
    3. Aliquot 1 µl (10pg -1 µg) of DNA to the chilled microcentrifuge tubes on ice.
    4. When the cells are thawed, add 50μl of cells to each DNA tube on ice and mix gently by tapping 4-5 times. For the pIG7spe control, add 1 µl of (500 pg/µl) DNA to the 50 µl of cells on ice. Mix well by tapping. Do not pipette up and down or vortex to mix, this can harm cells and decrease transformation efficiency.
    5. Keep tubes on ice for 5 minutes, and then transfer to liquid nitrogen for 5 minutes.
    6. Incubate tubes for additional 5 minutes in 37°C water bath.
    7. Immediately add 950µl of Recovery Medium or any other medium of choice to the tube, pipette up and down three times to re-suspend the cells.
    8. Incubate tubes at 30 °C for 3 hours at 200 RPM.
    9. Dilute the cells as appropriate then spread 20-200 μl cells onto a pre-warmed selective plate. For the pIG7spe control, you may plate 100 μl of undiluted transformation mix onto a YT plate containing 100 μg/ml spectinomycin. Use a sterilized spreader or autoclaved ColiRoller™ plating beads to spread evenly.
    10. Incubate the plates for 2 – 3 days at 30 °C.
